I'm growing rice plants hydroponically for a research project, and I was directed to this video (https://www.youtube.com/watch?v=ltbdM3boWmU) to use as a guide for germination prep. I've never made agar before, so could someone help make sure that my below protocol is correct for making agar that is safe for my plants? What type of agar powder should I be using? Furthermore, since my water solution will contain fertilizer, is it necessary that my agar contain nutrients? If not, do I just need to mix my agar powder with water? Thank you for all the help!

Protocol for 0.6% Agar:

1.) Add 4.31 g of MS Salts to 1.8 L of distilled water and stir to dissolve.

2.) Check and adjust pH to 5.7. Adjustments can be made with 1M KOH.

3.) Dilute to final volume of 2 L and add agar (10 g / L).

4.) Autoclave 15 minutes at 15 psi, 121oC.

6.) Optional sucrose and vitamins should be added after agar media cools, before pouring solution into a container (e.g. petri dishes, Magenta boxes, culture tubes).

you can use bacteriological or tissue culture agar.

if you are adding MS salt (Murashige and Skoog salt) medium then no need to add more vitamin and fertilizer.

to adjust the pH you should use 1M KOH and 1M HCl.

you are making 0.6% agar; then use 6 g Agar per liter.

if you want to add sucrose or different nutrient and vitamin then you can add when media become cool or when the medium temperature around 45oC.

You should use 200 ml distilled water for mixing MS salt after that you can make volume 2L.
