A student is interested in the transcriptional regulation of BOG1 (the bogum synthetase gene). Northern hybridization analysis shows that BOGI is strongly expressed in cells if Zubrium (Zub) is absent in the media and is turned off when Zub is present. She constructs a lacz fusion gene with BOGl (i.e. BOGI-lacz fusion gene; please refer to Figure 2 below), measures for lacz (B-galactosidase) activity and observes a pattern of expression of the lacz reporter gene in the presence and absence of Zub that is similar to the Northern hybridization results. She then decides to make site-directed mutations in three sites of the BOGIgene promoter (A, B and and then tests for lacz expression. Mutant sites are highlighted in black boxes in Figure 2 below. She also individually clones a wild type copy of each site, A, B and C into heterologous reporter promoters with and without the "upstream activator sequence enhancer" UAS) (please refer to the lower panel diagrams in Figure 2) and then measures lacz expression.

Explanation / Answer

Answer A) The region A is an activator. This activates the LacZ thus its response is more in presence of Zub in comnparison to the wild type. It is also inducible since it induces the activation of LacZ even in presence of Zub.

Region B is similar to region A. it is an activator as well as inducible.

Region C is constitutive. the transcription process of Lac Z in presence and absence of Zub is constant.

One can do more research on region C. to search if some changes can make the Lac Z to express more even in presence of Zub. She can try mutate the gene at region C by some other gene.
