Question
You perform column chromatography on a mixture of reaction products which shows two spots on TLC. However, under the column conditions you chose those two compounds do not separate. a. One can alter a number of factors in order to improve the resolute of components by column chromatography. For each of the factors listed in the following table, separation can be improved by either increasing or decreasing the factor. For each, describe the changes in sample size or interactions between components and the chromatography medium are altered by increasing or decreasing the factor so that the resolution will be improved. 1. Column Diameter: 2. Column Length: 3. Sample Volume: 4. Sample Concentration: 5. Loading Time: 6. Flow Rate:Explanation / Answer
flow rate , concentration and column diameter. TLC is a simple, quick, and inexpensive procedure that gives the chemist a quickanswer as to how many components are in a mixture. TLC is also used to supportthe identity of a compound in a mixture when the Rf of a compound is comparedwith the Rf of a known compound (preferrably both run on the same TLC plate).A TLC plate is a sheet of glass, metal, or plastic which is coated with a thinlayer of a solid adsorbent (usually silica or alumina). A small amount of themixture to be analyzed is spotted near the bottom of this plate. The TLC plateis then placed in a shallow pool of a solvent in a developing chamber so that onlythe very bottom of the plate is in the liquid. This liquid, or the eluent, is themobile phase, and it slowly rises up the TLC plate by capillary action.
