You are working in a laboratory on a project related to a protooncogene (PO). The mRNA of the gene is shown on the right. It is 550 nucleotides in length. There are 5 Hindlll sites on the shown sequence at nucleotides: 100 (1), 200 (2), 230 (3), 300(4) and 400 (5) (indicated by filled arrows). You know that there is a polymorphism (a SNP resulting in a missense mutation which makes this gene an oncogene (OG)) at the 3^rd Hindlll site which results in the loss of the Hindlll digestion site. You want to determine if 3 different patient tumors (A. B and C) contain this polymorphism, or not, by performing RFLP. You therefore, design two PCR primers (shown as open arrows on the sequence) and perform RFLP analysis. Draw the bands in the jel image above, which would indicate that tumor A contains no mutations. B is homozygous for the mutation and C is heterozygous for the mutation. Molecular weight markers (M.W.) are (from top to bottom) 500nt, 400nt, 300nt, 200nt and 100nt.
