The answer would be C) isoleucine with a valine. They arevery similar in size and chemical properties. A) tyrosinewith a glycine would cause a dramatic change. Their are a number ofreasons for this. You going from a hydrophilic large amino acid(tyrosine) to a very small hydropobic amino acid (glycine).Usuallly aromatic amino acids are very important for thefunctionallity of the active site of the protein because they arevery good at stablizing charges. Replacing the tyrosine with aglycine make also cause a change in the 3-d structure of theprotein do to the size difference glycine is very good at being inthe middle of radical bends and twists. It make also cause ahydrophobic interface on the surface of the protein which may causethe protein to aggerate together similar to the mutation in sicklecell. Answers B D and E all have similar reasons to my explainationto Tyosine and glycine.
