Q1. Fibroblasts placed on thin sheets of silicone rubber may exert tension and stretch the rubber to cause wrinkles. Explain how the ability of fibroblasts to wrinkle rubber would compare with that of normal cells under the following conditions.
a) The cells are injected with large amounts of purified gelsolin
b) The cells are treated with taxol
c) The cells are treated with latrunculin A, the drug is washed out, and the cells are observed periodically.
d) The cells are treated with latrunculin A, the drug is washed out, and the cells are observed periodically.
Explanation / Answer
Certain drugs act on the depolymerisation of micro filaments affecting the addition of G-actin to microfilaments.
a) When the cells are injected with large amounts of purified gelsolin…………..depolymerisation by changing the cross linking of actin filaments could be observed............This is the TRUE statement for the above given question.
Gelsolin is an actin regulator. It can assemble and disassemble the actin filaments. The name itself says that it converts a gel into a sol by disturbing the actin cross linking proteins.
b) The cells are treated with taxol………..............The toxin is either a protein or peptide or molecule causes the disease by contact.
c) The cells are treated with latrunculin A/B, the drug is washed out, and the cells are observed periodically...................latrunculin A & B are toxins, which disturbs the microfilament organisation especially addition of actin to the microfilaments of cytoskeleton. They maintain their altered state even after the washout of toxin.
