A child has a developmental disorder that appears to be the result of altered SHH signaling in the developing hand. Two models for this altered SHH signaling are: (1) The regulatory region of the SHH gene has been altered such that the level of SHH transcription was lower than normal in the developing limb bud (but not in other tissues), changing the shape of the SHH gradient. (2) There is a structural change in the SHH receptor pathway in the limb bud cells such that the SHH signal was normal but was not properly received or processed by the limb bud cells.

a.) Describe the experiment(s) you would do to determine which of these is correct.

b). Include in your answer the results you would expect from your test(s) if the first hypothesis is correct and explain why these results are convincing.

Explanation / Answer

a) Take a DNA chip for the given organism.

Then isolate the mRNA from the limb bud and normal tissue.

Hybridize the mRNA isolated from the two samples.

If the fist hypothesis is correct then the sample shows low fluorescence in DNA chip. So, it concludes that mRNA expression is low in sample 1.
