Briefly describe this products workings and uses, and claimed advantages.

The product brochure says: “The PureFection reagent self-assembles nanoparticles in the presence of DNA and RNA. These complexes are readily taken up by target cells for efficient gene delivery.”

Some speculation on your part (or find something in the literature)… Why are such self-assembled complexes (more?) readily taken up by cells than just naked RNA or DNA? I’m wondering.

Purefection technology uses nanoparticle based DNA delivery system. The company has its own nanoparticle which they are using for deliveriung DNA, RNA. The usage of this product very convenient for high throughput transfection experiments. The method involves easy-to-use protocol with rapid, one-step incubation for 15 minutes before adding directly to target cells.

Advantages include works with most cell types, nano-based gene delivery with low toxicity profile, rapid 15 minute protocol, works with both Plasmid DNA and siRNAs and cost-effective alternative to lipid-based products.

Efficient gene delivery, fundamental to the success of gene therapy remains a complex process with many possible rate-limiting steps. The most serious intracellular barriers are lysosomal degradation, nucleolytic degradation in the cytosol and inefficient delivery to the nucleus, whereas, extracellular barriers include nucleolytic degradation in the serum, recognition by the reticulo-endothelial system and nonspecific delivery.

Usually the naked DNA carry a negative charge and entry of the DNA is restricted into the cell due to the lipid nature of the cell membrane. Only neutral molecules or lipid molecules can pass freely through the membrane. Thats why DNA or RNA should be conjugated to a liposomal carrier for the efficient delivery into the target cells.
