bio genetic practice assignment, I know this a lot but any help would be nice!!

Question: For each panel of figure A-D describe the data depicted in that panel and the conclusions that can be drawn from the data.

I don't necessary need help with the conclusion part but what confuses me is understanding what is going on in each data, I understand in panel A that since H3K4me1 is euchromatic and ctrl KD is untreated Ash protein, the expression is normal and in the other two it is decreasing because they are treated with siRNAs that reduces expression but I don't understand what is going on in panel B, why is expression increasing if H3K9me3 is hetero chromatic and both ash21 kd1 and kd2 deals with low expression? also I don't understand what is going on in panel C and D, I know this seems a lot but any help would be much appreciated. also to note in panel C the very fist box is covered with green color.

itatos dilett transcription. This data analysis assignment primarily involves two such modifications: trimethylation of histone H3 lvsine residue 4 (H3 K4me3), and trimethylation of histone H3 lysine residue 9H3K9me3 histone H3 with just one or two methyl groups on lysine 4 (H3K4mel and H3K4me2 is similar to H3K4me3 H3K4me3 is almost always associated with active gene expression, in so called euchromatic DNA, while H3K9me3 is almost always associated with inactive or "heterochromatic The figure shown on the next page is adapted from a research article entitled "The Trithorax Group Protein Ash2l Is Essential for Pluripotency and Maintaining Open Chromatin in Embryonic Stem Cells", published in 2014 in the Journal of Biological Chemistry. It reports the results of experiments in which mouse embryonic stem cells were transfected with (took up) small inhibitory RNAs (siRNAs) that reduce expression of a protein callerash2ll a histone methyltransferase specific for histone H3 lysine residue 4, le, it is the enzyme that "writes' methyl groups onto H3K4, The experiments involve western analysis (panels a and b), in which antibodies are used to detect how much of a specific protein is present, in this case to determine how much H3K4me1, H3K4me2 H3K4me3, H3K9me3, and total histone H3 are present in embryonic stem cells that are treated in different ways. A western blot is like a southern blot, but detects specific proteins, not DNA fragments. They also involve a method called immunolocalization (panel C). in which antibodies are used to visualize at the cellular or tissue level how much of a protein is present, and where it is located. In this case one antibody detects a protein called HP1 which is associated with ranscriptionally inactive chromatin, or heterochromatin. A second antibody detects Ash21 DAPT is a dye that stains DNA.

Explanation / Answer

Expression of the Ash21 increases in hetero chromatic because expression of Ash21 treatment results in expression of ash21 kd1 and kd2.

In panel c the first column is green because it is a table of immuno histo chemistry where the fluroprobe for the protein HP1 alpha is green. Because the protein is expressed in all the three conditions it is appearing green.
