Question

You are studying the nuclear import of 3 different proteins by injecting purified, GFP- tagged versions into the cell cytoplasm. If you inject each protein individually, they are each rapidly imported into the nucleus. Next, you inject the proteins in pair-wise combinations with one of the proteins being fused to GFP and the other protein being unlabeled, and you obtain the results seen in Fig 1B and 1C. What would the results of 10 mu mol Protein L-GFP + 1mmol Protein L look like(Fig 1B or 1c)? Briefly justify your answer. What can you conclude about the import of Protein L, Protein Q, and Protein W? (Think "SIGNAL-RECEPTOR-TRANSPORTER")

Explanation / Answer

A. Protein L would enter the nucleus within 15 mins of injection. Various combinations of protein L fused with GFP as well as unlabelled protein show that in either case the protein resides within the nucleus within 15 mins.

B. Protein L and Q are cargo proteins which are imported to the nucleus wherease W appears to be a large receptor that helps in their import.Due to the heavy molecular weight of W (by itself as well as because of addition of a GFP tag) the movement becomes slow when W is tagged with these proteins L and Q.
