The theoretical concentration is always calculated by the means of estimation in cases of growth curves and cell counts. It does not take into the consideration of other environmental factors like availability of nutrients, accumulation of toxic substances and rate of error in the starting material. These concentrations are just a estimate calculations based on the numbers.

When you actually start doing the experiment, the concentrations will vary significantly when compared to the theoretical values. In the case of experimental setup many factors need to be considered for arriving at the right concentration. If you consider at the beginning of the growth curve, where cells enter a lag phase in which they acclimatize to the conditions for the growth. In theory we might consider that all the cells will divide, but experimentally this is not possible because some of the cells which are not able to adapt to conditions will evetually die and concentration of cells will differ with the theoretical value. The same in the case at the end of the growth, where toxic substances accumulate in the medium and retard the growth of the cells, so the experimental concentration will vary with the theoretical concentartion.

In short the environmental factors play a major role in experimental concentrations when compared to the theoretical concentration, where this factor is not considered at all.
