The following are errors or problems encountered in spetrophotometric applications. They can all be remedied by a change in instrumental or chemical conditions. State why each error or problem occurs and explain in a few brief sentences how the problem or error can be corrected.
(i) A UV-Vis spectrophotometric determination was found to have a precision of 2%, which was identical to the absorbance reading error on the strip-chart recorded readout.
(ii) In a biological kinetics study, once could be required to work at high absorbances (A = 3 to 4) to monitor a reactant in a reaction mixture. Dilution of the sample cannot be used because it changes the reaction mechanism. It is noted that absorbance is nearly independent of concentration at these absorbances.
