Need help with #17. Thank you!
BICH 410 F 2017 Assignment 8 60 points Due at the beginning of Class Friday, Nov. 10th 15.) (2 pts) If a relaxed circular double-stranded DNA molecule containing 874,500 bp in the B-DNA conformation were reacted with a DNA gyrase which introduced 17,674 negative supercoils into this DNA molecule, what would be the values of L, T and W? 16.) ( 2 pts) If a relaxed circular double-stranded DNA molecule containing 874,500 bp in the B-DNA conformation were reacted with a DNA gyrase which introduced 17,674 negative supercoils into this DNA molecule, what would be the superhelical density, ? 17.) (2 pts) How many ATP molecules would be required for DNA gyrase to generate 17,674 negative supercoils?Explanation / Answer
Ans. DNA gyrase is heterotetrameric (A2B2) enzymes involved in creating and relaxing supercoils.
The enzyme has one ATPase domains that binds to and hydrolyzes ATP to create a negative supercoil. That is, 1 ATP is hydrolyzed per negative super coil.
So, total number of ATP molecules required to generate 17674 supercoils shall be equal to the number of supercoils.
Hence, number of ATP required = 17674 ATP
