Question

Your laboratory has just gained the license to allow you to research, and commercialize the results for, a novel Protein 77(p77). Because there is very little known about this protein, you need to characterize the protein. You are provided with: the cDNA corresponding to the protein, a Chinese Hamster Ovary (CHO) cell line expressing the cDNA for the p77, and the amino acid sequence (using the one-letter code) of the p77.

Shown following is the first 30 amino acid region of this putative amino acid sequence for p77:

1YFNPT 6DSWPT 11LAPAN 16YSFLF 21FLARY 26WYINL30

Explain two approaches which would demonstrate that the cell line provided expressed the cDNA construct for your protein.

Explanation / Answer

* Single cell gene expression analysis: Single cell transcriptome analysis through fluorescence imaging. Through live imaging, without degrading the cells or cells of interest, the target protein expression can be analyzed. Moreover, localization of the target proteins also can be visualized.

* Fluorescence in-situ hybridization: a probe specific for the cDNA (target) is labelled with a suitable fluorochrome, and used to analyze the target cell.

* PCR based quantification of mRNA
