Explanation / Answer

It is the condition utilized for HDV transfection of mammalian cells. The RNA polarity is positive for cDNA mediated transfection and negative for RNA mediated transfection. cDNA mediated transfection is sensitive to a-amanitin and it requires small delta antigen and the polymerase is RNAP II and the site of synthesis is nucleoplasm, where as in RNA mediated transfection it does not require small delta antigen and it needs RNAP I and the site of synthesis is nucleolus.
