Question
19) In our experiments in the laboratory we packed the column and eluted the spinach extract. We packed to column by making slurry of the silica gel in petroleum ether. Is it a wet packing or a dry packing? 20) What experimental technique we use to separate a mixture of high boiling liquids? 21) Apart from silica gel, which other compound can be used as a stationary phase preparative column chrarmatography? 22) If silica gel is the stationary phase in preparative and thin layer chromatographs, what is the mobile phase? Is it the solvent used for driving the compounds or the compounds themselves? 23) What is the purpose of putting sodium sulfate (NaSO) crystals in the organic layer isolated after extraction? 24) Can we use a mbxture of solvents for eluting compounds in TLC and Preparative Chromatography? 25) What are the similarities and differences between Thin layer chrometography (TLC) and Preparative Column chromatography? Explain.Explanation / Answer
19. The slurry of silica gel in petroleum ether is used for wet packing. In this case the column is packed with a slurry of the stationary phase(silica gel) in petroleum ether. So in this case it is wet packing.
Note- in case of dry packing , the silica gel powder is first poured into the column. This is followed by adding petroleum ether into the column.
