3. Problem 8.6 (from text book) 8.6 a. Describe the similarities and differences between two types of HPLC: size-exclusion chromatography and adsorption chromatography. b. Assume the graph below was produced using reversed-phase adsorption chromatography. One of the curves represents a polysaccharide (sugar), while the other represents the same polysaccharide covalently linked to poly(styrene) for a drug delivery application. Which curve is associated with which and why? Explain in terms of molecular interactions with the stationary phase of the column. 12 T 10 0 O214 Elution tme (min c. Now assume that the graph above was produced using gel (a type of size exclusion chromatogiaphy) for the same molecules as described above Which curve is associated with which and why Explain in terms of molecular interactions with the stationary phase of the columnExplanation / Answer
a) Answer:
similarities
Differences

b)
As we know that typical reversed phase stationary phases are hydrophobic and chemically bonded to the surface of a silica support particle and the stationary phase is hydrophilic in nature. So, molecules with hydrophilic properties contained within the mobile phase will have a high affinity for the stationary phase.
In our case, polysaccharide(sugar) is a more hydrophilic in nature then polysaccharide covalently linked to poly(styrene). a graph which has less elution time is representing polysaccharide (sugar) and a graph which has more elution time is representing polysaccharide covalently linked to poly (styrene).
--------------------------------------------------------------------------------------------------------------------------------------------------------
c) size exclusion chromatography related to molecular weight, so molecular weight of polysaccharide covalently linked to poly (styrene) is more than a molecular weight of polysaccharide (sugar).
Molecules which have more molecular weight has less elution time i.e. molecular weight is inversely proportional to elution time. Hence a graph first is representing polysaccharide covalently linked to poly (styrene) and graph second is representing polysaccharide (sugar).
