Question

Isolation of Eugenol from Cloves objectives Learn how to carry out a steam distillation. Separate components of clove oil via acid-base extraction. Characterize eugenol using IR and NMR spectroscopy. Determine purity of components using TLC analysis. OCCH3 OCH OCH3 CH2CH CH2 CH2CH CH2 acetyleugenol eugenol Adapted from Williamson, Kenneth L and Masters, Katherine M., Macroscale and Microscale organic Experiments, 6th edition. Belmont, CA: Brooks/Cole, 2011, pp. 111-113. Reference Material The PowerPoint presentation for this experiment is provided on eCampus. Molurig: pp 194-196. A simple distillation setup (Figure 12.7 on p 181) will be used. Procedure Approximately 5 g of whole cloves are weighed into a beaker and transferred to a 250 mL round- bottom flask with a thermometer inlet. A few boiling chips and 100 mL of water are added to the flask. A simple distillation setup is constructed, and the Powermite is set to 45% of full power. A 125 mL Erlenmeyer flask is used to collect the distillate. (Make sure to use a flask that has a

Explanation / Answer

1) safecty precautions : wear the following protective equipment when using HCl or NaOH of any concentration:

-Vapor respirator
-Rubber gloves
-Boots
-aparon/ lab coat
-Face shield mask
-access to an eye-flush area in case of accidental exposure.

-Always add corrosives slowly to cold water.

-Keep containers tightly closed when not in use or empty.

- Store in an area that is: well-ventilated, cool and dry place separate from incompatible materials.

2) steam distillation is the method used for separating/isolate substances which are having high vapour pressure at the boiling temperature of water, immiscible with water and steam volatile.

And Eugenol and acetyleugenol both have above characteristic. So, steam distillation is the preferred method.

3) first dissolve the mixture into dichloromethane

- then add aqueous solution of base to abstract proton from eugenol and forms salt with it.

- then shake the mixture for 10 minutes and kept it for sometime to allow layers separation.

- the layer containing eugenol is collected and washed with Saturated solution of Nacl in separatory funnel.

4) This is done for :

- drying process ie to remove water that may have dissolved in the organic phase.

- prevention of emulsion formation.
