Assuming that a concentration of PNPP to obtain 1/2 Vmax of Alkaline Phosphatase is 15 mM, calculate its Km in the presence of a noncompetitive inhibitor? After calibrating your column with different protein of specific molecular weights, you determine that the straight line formula to be Y = -0.5X + 30. Calculate molecular weight of your protein of interest with V_e of 10 ml. When you increase temperature of your enzymatic reaction from 25 degree C to 37 degree C, the rate of the reaction would increase. Would you expect to continue seeing the increase in the rate of reaction as you gradually increase the temperature? Explain.

5) In this experiment , we took a tube of pNPP without any enzyme because pNPP is colour less. so it will the initial observation of pNpp. when you are adding enzymes with increasing concentrations, the clour will slowly changes .

Addition of enzyme leads to pNPP to pNP . which is bright yellow colour.

(8) The rate of reaction is very high for enzymes in the temperature range of 35 - 40 oC , if you increase further more than this, the enzyme will inhibit or die as they are biological species which lead to decrease in the overall reaction. So, the optimum temperature will be around 36- 38 oC
