This is an extra credit assignment I have done. We have not discussed these topics thoroughly in class so I would love to double check what I have done. The amino acid polypeptide chain is GKDFQKKAVSLHCGC. I have already drawn this and feel confident with it.
1) Predict the overall charge ay pH 4.62 and pH 9.95. (I use the table method for clarity) MY ANSWER: +2, -1
2) Determine the pI and reason why MY ANSWER: 8.89, b/c the neutral N terminus pka = 9.60 and the cysteine R group pka = 8.18, average the two
3) assume that this peptide sequence represents the sequence of amino acids found in the active site of an enzyme. The enzyme only functions when the charge of the active site is -4. Would this active site function at a normal physiological pH of ~ 6.8. Explain your answer. No, b/c the overall charge would be +1, this is too acidic for this active site.
