Question

100 mg of ibuprofen has mixed with 25 mL octanol and 25 mL water inside a separating funnel in order to determine its partition coefficient. After aqueous and organic phases are separated, determination of absorption of aqueous phase and construction of a calibration curve using UV-spectrophotometer has been applied. Concentration of ibuprofen in aqueous phase (water) is found to be 0.0125 mg/mL.

If the dissociation constant (Ka) of this drug is 3.02 x 10 -5 , after taking the pill orally, where will this drug be absorbed across membrane to reach the liver and bloodstream? Justify your answer.

Explanation / Answer

As we can see from concentration of ibuprofen in aqueous phase. It is not soluble there.

Ibuprofen is absorbed in stomach and at low pH 2-6.

Acidic drugs are absorbed faster in acidic pH as they remain unionized in acidic medium of stomach. As ibuprofen is soluble in organic layer it is absorbed through lipidic cell membrane
