Question
A cloned fragment of DNA was sequenced by using the
dideoxy chain-termination method. A part of the autoradiogram
of the sequencing gel is represented here.
a. Deduce the nucleotide sequence of the DNA nucleotide
chain synthesized from the primer. Label the 5'
and 3' ends.
b. Deduce the nucleotide sequence of the DNA nucleotide
chain used as the template strand. Label the 5'
and 3' ends.
c. Write out the nucleotide sequence of the DNA double
helix (label the 5' and 3' ends).
d. How many of the six reading frames are ?open? as
far as you can tell?
I'm really having trouble with part D
Explanation / Answer
a.) The nucleotide sequence of the DNA nucleotide chain synthesized from the primer :
5'--TTCGAAAGGTGACCCCTGGACCTTTAGA--3'
B.) The nucleotide sequence of the DNA nucleotide chain used as the template strand :
3'--AAGCTTTCCACTGGGGACCTGGAAATCT--5'
c.) The nucleotide sequence of the DNA double helix (label the 5' and 3' ends) :
5'--TTCGAAAGGTGACCCCTGGACCTTTAGA--3'
3'--AAGCTTTCCACTGGGGACCTGGAAATCT--5'
d.) Open reading frame in known as a frame that can be transcribed in mRNA and translated in a protein. It starts with Start codon and stops with stop codon amd the region between start and stop codon expresses any protein. I could be done through any online free software like ORF finder, ORF invetigator, ORF predictor etc.
