Dideoxynucleotides causes chain termination. Of the four nucleotides, each one is added at each step. A dark band indicates DNA fragment that is the result of chain termination after incorporation of a each dideoxynucleotide (ddATP, ddGTP, ddCTP, or ddTTP). The DNA sequence then can be read base on the relative positions of the different bands from bottom to top.
