Rubisco-PAGE Lab question help!
2. What is/are the differences between the chloroplast sample(C) and your total protein sample ("T")?
3. What is/are the differences between your chloroplast sample and your soluble sample ("S")? As you did for question 3, provide an explanation for any differences or lack thereof.
T=5.0 mL of Isolation Medium,5 grams of grinded spinach leaves,
C= whats left on bottom after centrifugation(This process pellets any additional chloroplasts and other organelles, leaving only soluble (i.e. cytoplasmic) protein in the supernatant. ) of sample T.
S=supernatant from C
Can you help me with these questions? Q..Q
Explanation / Answer
2. The total protien sample (T) contains the spinach leaves and an isolation medium. Upon centrifugation the chloroplast and some cell organelles settle down as pellet. Therefore the Total Protein sample (T)contains all cytoplasmic proteins along with the cell organells but Sample (C) specifically contains chloroplast
3. Chloroplast sample is pellet formed after centrifugation of Sample T whaereas, Soluble Sample is Supernatant obtained from centrifugation of Sample T. The sample C contains the chloroplast from spinach leaves and some cell organelles whereas the sample S contains the soluble, cytoplasmic protein.
Chloroplast sample settle as pellet because it is heavier than the soluble protein.
