Question

a sample has a density of 1.37 X 10^5 CFU/ml.
What sample volume should yield a countable plate?
Which two dilutiom tubes could be used to produce this sample volume? how?

i know the equation for density (ocd) is
OCD=CFU/DV

and what is considered countable plate?
number of colonies under 30 is considered countable?

and in this question what does it mean bt "which two dilution tubes"?

i found this question asked by another person with an answer but couldnt understand.

please explain the answer step by step.

thank you

Explanation / Answer

Answer:

The sample volume of 100 microlires or 0.1 ml, will yeild good plate count

The duplicate plating or triplicate plating is done for same sample and same volume, for better results and average of the duplicate or triplicate are taken for final CFU/ml(colony forming units).

The colonies less than 200 cfu, in a plate is considered for counts. if plate has 30 colonies, it is best suitable for counting
