2 Explain the action of pancreatic lipase on its substrate ( which are the products ? ) using the data from the lipase experiment explain how bile and temperature affect the activity of lipase.

The pancreas produces pancreatic juice that is a clear and composed mostly of water along with some salts, sodium bicarbonate, and several digestive enzymes such as amylase lipase, nuclease and other protein-digesting enzymes.

Pancreatic lipase produced by Pancreatic acinar cells and act on triglycerides ( lipid / fats) which have been emulsified by bile salts and break down them into fatty acids and monoacylglycerides.

Bile is produced by liver. It is a yellowish alkaline liquid comprises water, bile salts, bile pigments, phospholipids , electrolytes. It is further secreted into the small intestine, where bile salts present in it emulsifies fats and thus it provides a larger surface area for pancreatic lipase activity results in an increases the digestion of fats. Thus if there is lipase test with no bile ( test tube 2) it significantly reduce the activity of lipase enzyme.

Temperature also affects the activity of pancreatic lipase as increasing temperatures (up to 40 ºC) increases the collision rate between the lipase and triglycerides molecules and thus increase lipase activity. Thus for test tube 3 incubated at 37ºC , there is high lipase activity than in test tube at room temperature ( 27ºC) .

3 What is bile ? Where is it produced ? Explain it’s function in digestion.

Answer:- Bile is a yellowish alkaline liquid comprises water, bile salts, bile pigments, phospholipids and electrolytes secreted by the Hepatocytes of liver. Components of bile acts on large lipid molecules emulsify them and thus increase the surface area for the activity of pancreatic lipase which breakdown triglyceride lipids into fatty acids and monoacylglycerides that easily get digested. Thus how bile helps in digestion process.

4 Where in digestion tract do lactase and pancreatic lipase usually function? What would be the consequence of a deficiency in either of these digestive enzymes ?

Lactase is produced in small intestine where as pancreatic lipase is secreted in pancreas. Both of these enzymes usually function in small intestine of digestive tract. The functions belongs to lactase is the break down of lactose ( disaccharide) into one molecule of glucose and one molecule of galactose( monosaccharides). The deficiency of lactase can lead to lactose intolerance where as lipase act of fats and break down them to simple products to digest , the deficiency of pancreatic lipase leads to fat malabsorption that can cause various disorders like pancreatitis and bowel diseases.
