Question

3. In a bright-field compound microscope, starting from the illuminator, follow the pathway of light: a. The first lens that provides magnification of the image is thevlens b· The lens that further enlarges the image ten more times is the lens. 4. Open the iris diaphragm all the way while using the low power objective. What happens to the field of view and the quality of the image? Now close the iris diaphragm all the way. What happens to the field of view and the image? Why should you always begin with scanning or low power objective? And why you should not begin with high-dry lens? 5. 6. Why were you asked to use only the fine adjustment knob after switching to 40X? 7. Complete the following table to calculate total magnification for the compound microscope: Objective Objective Lens Ocular Lens Power Total Magnification 40X Lens Power 4X iox Scanning Low power High-dry Oil immersion 8. What is the highest total magnification that can be achieved using the bright-field compound microscope?

Explanation / Answer

4. Iris is an aperture that controls the light that falls on the specimen. If you completely open the iris the light intensity inceeases and one may strain their eyes and may wash out the image completely. If you completly close the iris the light intensity would be 0 and the image won't be visible any more.

5. One should always start the low power lense because one has to locate and get an overall structure of the specimen before one moves on to high powered lense to observe the specimen at higher magnification. ,It's easier to study the specimen with this strategy

6. 40x lense is longer and wider, if coarse adjustments are made then the lense will rise with longer motions which may break the slide and damage the lense. If fine adjustments are made then the lense would make small motions preventing any damage to the objective lense.

7. Low power 10x 10x 100x

High power. 10x 40x 400x

Oil immersion. 10x 100x 1000x
