Explanation: The RB (Retinoblastoma ) gene located on the chromosome 13q14 is normally present in the active state in every cell. It arrests the cell cycle at the late G1 phase. Various growth factors inactivate the RB gene by phosphorylating it which removes the G1 cell cycle block allowing the cell to pass from G1/S block point and to proliferate.

The p16INK4A Tumor Suppressor Protein functions as an inhibitor of CDK4 and CDK6; which can initiate the phosphorylation of RB. Thus p16INK4A protein can arrest cells in the G1 Phase and lead to cellular senescence (irreversible growth arrest). Cellular senescence plays an important role in tumor suppression and hence, suppressing the expression of p16INK4A protein will help a cancer cell to bypass its normal regulation of cell cycle by RB and lead to excessive proliferation by removal if the G1/S cell block.
