Question

Scientists are now able to reprogram adult, fully differentiated cells into more naive cells with the capacity to differentiate into multiple cell types called induced pluripotent stem cells (IPSCs). There are four markers that become expressed when the cell transitions from terminally differentiated to an IPSC: Oct4, Sox2, Klf4 and c-Myc. The average expression level of 0ct4 (sample mean) is 3472 normalized copies (extra info: normalized to another gene-GADPH). The sample standard deviation is 80 normalized copies. The sample number is 28 dishes. In each problem, place the finding in the context of the problem, (z-table and t-table available on Blackboard) 1. What is the 90% Cl for the true expression of Oct4 in IPSCs? 2. What is the 90% Cl for the true expression of Oct4 in IPSCs if the sample size is 7625 dishes? 3. Assuming a z-distribution, what percent of IPSCs express higher than to 3500 normalized copies of Oct4?

Explanation / Answer

Solution: Given Sample mean = 3472 , sample standard deviation = 80 , sample size = 28

a) We have given sample standard deviation, also sample size is <30, hence we will use t distribution for finding CI.

90% CI is (3446 , 3498)

b) Here we have given sample size 7625,hence we will use z distribution

90% CI is , (3470.49, 3473.5)

c) Here we have to find P( x> 3500)

Using excel, =1 - NORMDIST(3500,3472,80,TRUE)

We get P( x > 3500) = 0.3631
