You should know characteristics of the bacteria that grow on them such that you can ID the bacteria growing on a specified differential medium. Know composition of each type of media and what it means when bacteria grown on them (what is happening biochemically).

1.Gelatin test: Gelatin test is used to detect the ability of an organism to produce gelatinase. that liquefy geltatin. gelatinase degrade gelatin to polypeptides then polypeptides are further converted in to amino acids. The bacterial cellls can these cells take up amino acids and use these in their matabolic processes. Gelatin hydrolysis test is helpful in identifying and differentiating species like Bacillus , Clostredium and Protues.
2.Synder test: Synder test is used to determine a persons suseptability to dental cavities. the suspectability is correlated with acid production . that is assumed to result from fermatation by cariogenic lacto bacillus species on the teeth or in other area of the mouth. test agar contains 2% glucose , PH of the agar below 4.8 which inhibitsb the growth of most organisms, but it is ideal for acidophilics like Lacto bacillus.
3. Urease broth: Urease broth is a differenatial medium that tests for ability of an organism to produce an exoenzymes called urease. that hydrolyse urea to ammonia and carbondioxide. urea broth is formulated to test for rapid urease -positive organisms. the restrictive amount of nutrients coupled with the use of pH buffer prevent all , but rapid urease -positive organisms from producing enough ammonia to turn in to phenol pink.
4.SIM test: Sim medium is a combination of different media that tests for different parameters

if the organism can reduce sulfur to hydroxy sulfide, the hydroxy sulfide will combine with the iron to form ferric sulfide which is black precipitate, if there is any blacking of the medium . it indictes that reduction of sulfur.
