Section from article: AuNP–Pep1 was designed to bind specifically into the active cleft of HEWL (Fig. 2B) as cAb-Lys3 (Fig. 2A), so the inhibition of HEWL’s activity can be used to quantify the binding between AuNP–Pep1 and HEWL. Fig. 3A shows the activity profiles of HEWL after incubation with free Pep1 or AuNPs (3.6 nm) conjugated with different peptides. The decreased slopes represent the inhibition of HEWL activity due to the binding with different species. It can be seen that free Pep1 does not affect the activity of HEWL, indicating that free Pep1 does not bind to HEWL. It is unsurprising that the nonfunctionalized AuNPs could inhibit the activity of HEWL completely, because it is well-known that there is strong nonspecific binding between the nonfunctionalized AuNPs and proteins, forming so-called protein corona on the surface of AuNPs (27??–30).

Explanation / Answer

AuNP–60Pep1s has the similar arrangements of amino acid, also the similar density of peptide and stretch over, as AuNP–60Pep1; moreover, the Pep1's contended sequence remove the HEWL specificity. As wonted, simply between AuNP–60Pep1s and HEWL weak nonspecific interaction is noticed. For Pep1m, it has the similar sequence as Pep1 other than only one AuNP anchor point, and hence can migrate more freely on the surface of AuNP than Pep1. Because fixing the large flexible Pep1m to the binding arrangement will leads a great conformational penalty of entropy, AuNP–60Pep1m inhibiting the HEWL is only similar to that by AuNP–60Pep1s, much smaller than that by AuNP–60Pep1.
