Thanks in advance.**

FORM B MULTIPLE CHOICE. Choose the one alternative that best completes the statement or answers the question. 1) RNA polymerase and DNA polymerase differ in that 1) A) RNA polymerase uses RNA as a template, and DNA polymerase uses a DNA template. B) RNA polymerase binds to single-stranded DNA, and DNA polymerase binds to double-stranded DNA. O) RNA polymerase is much more accurate than DNA polymerase. D) RNA polymerase can initiate RNA synthesis, but DNA polymerase requires a primer to initiate DNA synthesis. E) RNA polymerase does not need to separate the two strands of DNA in order to synthesize an RNA copy, whereas DNA polymerase must unwind the double helix before it can replicate the DNA. 2) In trying to determine whether DNA or protein is the genetic material, Hershey and Chase made 2) use of which of the following facts? 2 A) DNA contains phosphorus, but protein does not B) DNA does not contain sulfur, whereas protein does. C) DNA contains nitrogen, whereas protein does not. D) A and B only E) A, B, and C 3) For a repressible operon to be transcribed, which of the following must be true? 3) A) A corepressor must be present. B) RNA polymerase and the active repressor must be present. C) RNA polymerase must bind to the promoter, and the repressor must be inactive. D) RNA polymerase cannot be present, and the repressor must be inactive. E) RNA polymerase must not occupy the promoter, and the repressor must be inactive. 4) Which of the following are nitrogenous bases of the pyrimidine type? 4) A) guanine and adenine B) cytosine and uracil C) thymine and guanine D) ribose and deoxvribose E) adenine and thymine

Explanation / Answer

Answer -

Question 1. The option (D) is correct.

RNA polymerase can initiate RNA synthesis, but DNA polymerase require a primer to initiate DNA synthesis .

Explanation - Because in case of DNA synthesis, DNA synthesis cannot initiate on a bare template strand.DNA polymerase require a nucleotide 3'- OH group and start adding new nucleotide. Primer provide a site for adding new nucleotide .So that, for initiation step DNA polymerase require primer.

But in case RNA polymerase no requirement of primers.

Question 2. The option( A) is correct. DNA contain phosphorus but protein not.

Explanation - In Hershey and Chase experiment, they use radioactive isotope technique ,They check movement of DNA and protein during phase infection. When result are coming they found radioactive phosphorous goes into progeny by reproduction. So that, DNA is genetic material. On the other hand no radioactive detect in protein phase(sulphur radioactive) .

Question 3. The option (C) is correct. RNA polymerase must bind to the promoter and repressor must be inactive .

Explanation - when the repressor (inactive) ,it unable to bind operator region. so RNA polymerase easily bind to promoter region and start transcription of structural genes.

Question 4. The option (B) is correct. Cytosine and uracil.

Explanation - because pyrimidine consist of six- sided ring only.
