QUESTION 9
(4 marks)
A fragment of DNA is cloned into a plasmid with a sequencing primer binding site. After dideoxy sequencing, the gel pattern shown in this diagram is obtained.
(a) What was the sequence of the complementary DNA strand in the sequencing reaction? 5' 3'
(b) What was the sequence of the template DNA in the sequencing reaction? 5' 3'
Please write your sequencing in the correct 5' to 3' direction. Deductions will be made for sequences input in the incorrect direction.
HINT: Consult Figure 14.14 in the textbook.

QUESTION 10
(3 marks)
DNA fingerprinting (or DNA profiling) is the process where a specific DNA pattern, called a profile, is obtained from a person or a sample of bodily tissue.
Most of our DNA is identical, however, specific regions are highly variable between individuals and differences in these regions are known as polymorphisms. Each of us inherits a unique combination of polymorphisms from our parents. DNA polymorphisms can be analyzed to give a DNA profile, with each of us having a one-of-a-kind DNA fingerprint.
Each of the following molecular markers can be used to generate a DNA profile or differentiate between individuals. Match each with the correct description.
Restriction fragment length polymorphism (RFLP)
Single-nucleotide polymorphism (SNP)
Short tandem repeat (STR)
A unit of two to thirteen nucleotides repeated hundreds of times in a row on the DNA strand.
Variation in the pattern of fragments produced when DNA molecules from different individuals are cut with the same restriction enzyme.
Single base sites that differ among individuals.
Restriction fragment length polymorphism (RFLP)
Single-nucleotide polymorphism (SNP)
Short tandem repeat (STR)
A.A unit of two to thirteen nucleotides repeated hundreds of times in a row on the DNA strand.
B.Variation in the pattern of fragments produced when DNA molecules from different individuals are cut with the same restriction enzyme.
C.Single base sites that differ among individuals.
Explanation / Answer
