2)You will determine the KM and VMAX of the uninhibited reaction, and the apparent KM and apparent VMAX of the inhibited enzyme. (DO NOT FORGET THE UNITS!)
3) Write a mechanism for each reaction and tell why the KM and Vmax change on the basis of the mechanism.

1) In the hyperbolic (Michaelis Menten) plot, the black line represents uninhibited and the blue line represents the inhibited reaction. In Lineweaver Burks plot, the straight line represents the uninhibited and the dotted line represents the inhibited reaction.
2) For uninbited reaction:
1/Vmax = 0.75, Vmax = 1.33 umol/sec
-1/Km = -1; Km = 1 umol
For inhibited reaction:
Vmax = 1.33 umol/sec
-1/Km = -0.5; Km = 2 umol
3) This is a competitive inhibition. Both the substrates inhibitor compete for the active site on enzyme. So, Vmax remains same and Km is increased.
