Question

A hypothetical plasmid vector called pYme is similar to pUC19 except pYme contains antibiotic resistance genes for tetracycline-resistance, chloramphenicol- resistance and ampicillin resistance. The pYme plasmid does not contain a B-galactosidase gene. Instead, the pYme plasmid contains a polylinker (multiple cloning sires) located in the open reading frame of the tetracycline-resistance gene there is a single HindlI another restriction endonuclease) recognition site in the polylinker of the pYme plasmid into which foreign DNA sequences (with Hindil compatible ends can be cloned. Suppose you wanted to clone Hindill fragments of vampire bats genomic DNA pYme vector d. You have completed the ligation reaction and are ready to do the transformation when a problem arises. Your fellow graduate student has told you that that the preparation of pYme vector DNA that you are using also contains significant amount of the pReCA/pUC19 recombinant plasmid. How would you redesign you transformation experiment to enhance your changes of recovering pYme recombinant clone containing vampire bat DNA while minimizing the chances of recovering the pRecA plasmid.

Explanation / Answer

there is a difference in antibiotic resistance gene between pYme and pUC19. pYme vector contains antibiotic resistance genes tetracyclin, amoxicillin and chloromphenicol. On the other hand pUC19 contains only ampicillin resistance gene. Thus after transformation if the bacteria colonies are plated on agar plate containing both tetracyclin and chloromphenicol antibiotic, plasmid which only bacteria which only contains pYme with the vampire bat genomic DNA will survive and bacteria with pUC19 can not grow.
