Most of the caspases play an important role in the apoptosis, which is described as programmed cell death.

The activation of the caspases involves the stages namely, dimerization and cleavage.

The dimerization process is facilitated by the binding to the adaptor proteins.

This binding is through the protein-protein interaction. Some of the activating multiprotein complexes formed during the caspase activation are:

Inflammasome

Apoptosome

Death-inducing signaling complex.

In the cleavage stage, at the inter domain linker regions, the caspases cleave. This leads to the formation of small and large subunits. It helps the active-site loops to take the information which is favorable for the enzymatic activity.
