Question
6. After graduating from undergrad. You decided to become a graduate student in Dr. Peach lab, who is known to utilize a variety of techniques (which you focused on in biochem 442) and has unlimited grant money (yeah she's ballin). You are interested in a protein named Mario. Mario is known to interact with three different proteins (called Luigi, mushroom, and feather) which are expressed in Yoshi cells. Dr. Peach tells vou that she has identified a mutant version of the Mario in a Yoshi cell line, but is not sure whether this mutation affects the ability of Mario to interact with the other three proteins. Your task is to test whether this mutation can affect the interactions between Mario and the the other proteins, You will need a lot of the Mario protein to do the in vitro assay, therefore first you will need to purify mutant Mario protein in E.coli. While doing this, Dr. Peach wants you to mentor two undergraduates (named Kirby and Navi) who have never done research before. Write a protocol for your undergraduates on how to purify protein. Start with isolating mutant mRNA from Yoshi cell lines. You can use any technique that was mentioned in class! You can also use combinations (ex: qPCR then sequencing) as long as you explain why each technique is necessary to answer that particular problem.Explanation / Answer
In order to isolate the protein of interest from the bacteria, following simple procedure can be followed:
