The Hfr strain is prototype which utilises the sugar lactose i.e, it has gene for synthesis of lactose. So if we supply glucose in the media and if there is a growth of bacteria this says the bacteria produces lactose. So we can add glucose where it produces lactose from that and determines lactose producing organism.
Generally the F+ plasmid contains marker genes tyr and str, so only tyr is transferred to exoconjugate. To check whether the atr gene is transferred the bacteria is grown with supplement of streptomycin. If growth occurs, states transfer of str gene and if no growth indicates no transfer of gene str.
The master plate include lysine, its already stated that only tyr will transfer to Hfr strain. So we have to supplement master plate with lysine.
