Answer 1: Nissl-stain is an organi-acid based stain which gives bright blue and pink color to the tissue. Thus, in order to permanently seal the tissue-slides stained with Nissl-stain, the slides/coverslips are often mounted in permount as a medium of choice.

Answer 2: Since permount is a stain which permanently seals the glass slide but cannot prevent photo-bleaching of fluorescent dyes, it is not mandatory to mount a DAPI-stained tissue in permount. On the contrary, permanents mounting media are commercially available which prevent photo-bleaching.
