Question

A and D are incorrect. Why is D incorrect? What is the correct answer? b. Asparagine and glutamine resid into aspartic acid and glutamic acid, respectively. If this happens to a p protein, it will result in a mixed population of proteins with the original Asn and GIn resides and proteins in which they have been replaced by ASp and Glu. What would be the best technique to separate original wild-type proteins from th deamidated ones? urified A) SDS-PAGE S2 B) Solubility assays (salting out) Gel filtration D) Jon exchange be Asp and Gl are chaaua harae Ineutra poin

Explanation / Answer

Why D is incorrect? - Ion exchange seperation based on the net charge of proteins and deamidation may or may not affect the net charge so this process cannot be appied for seperation of wild type and deamidated proteins.

Correct answer - C) In a protein deamidation may alter its structure, stability or function and may lead to protein degradation. The resulting mass cahnge is +1 Da shift. So the seperation of wild type from deaminated protein can be simply done by gel filteration. As because of the degradation the compact size of the protein becomes larger and hence can be checked by gel size exculsion filteration.
