Question
a.2 c.3 d.5 e.The answer cannot be predicted. Experimental approach (50 points) You purchased a batch of genetically engineered Bt-sweet corn hybrid seeds from the company which claimed that the seeds are genetically altered to express one or more proteins from the bacterium Bacillus thuringiensis Delta endotoxins, which is poisonous to certain insect pests and is widely used in organic gardening. Design an experiment in detail to identify if the seed are actually genetically altered (20 points) 1-Explanation / Answer
We can perform various tests to check whether the given seeds are genetically modified or not.
i. We can use PCR to check for the presence of the cry gene sequence that codes for the endotoxin. (We can also perform Southern blotting to check whether the given seeds contain the respective genomic sequence).
ii. We isolate RNA from these seedlings and perform RT-PCR to identify whether the transgene is being expressed. (We can also perform Northern blotting)
iii. We can isolate proteins from these seedlings and perform western blotting with endotoxin specific antibody.
iv. We can grow these plants in the presence of the insect and check for its resistance/sensitivity.
