Question

2) (3 pts) Listed below are the base compositions of three different duplex DNA molecules I. 5' ATTCTAACTTTGAT 3 II. 5 CGCACTGGCTAACC3 II 5' CGCATTATTGTCAA 3 3 GCGTGACCGATTGG 5 3 TAAGATTGAAACTA 5 3 GCGTAATAACAGTT 5 a) Order these three molecules in terms of their melting points from highest to lowest. b) State the relationship between the percentage of AT base pairs in a double- stranded DNA molecule and the melting point of that molecule c) Order these three molecules in terms of their density from highest to lowest.

Explanation / Answer

DNA I) melting point= 25.7

DNA II) meltig point = 43.2

DNA III) melting point= 31.5

a) So the melting point of II DNA is highest and I DNA is lowest. IIDNA > III DNA > I DNA.

b) The melting point of a DNA with high AT percentage have low melting point compared to the one with high AT percentage. This is because the GC forms three hydrogen bonds whereas the AT forms two hydrogen bond SO, a DNA with more AT will have lower melting point.

c) The density of a DNA depends on its GC content the higher is the GC content the denser is the DNA. So based on density the above three DNA can be ordered as IIDNA > III DNA > I DNA.
