On The DOG gene is expressed in cats and is regulated by several proteins, two of which -term ter of te 06 gre are termed protein 1 and protein 2 These bind to regions in se gene termed region 1 and region 2 A schemaltic of the DOG gene is shown below 2 REGULATORY PROTEINS BINDING SITES 2 3 3" START SITE FOR THE DOG GENE To determine how these proteins control transcription of the DOG gene, you isolate the gene from various cats and determine the DNA sequences of the binding sites you also measure the level of DOG mRNA produced in these cats' cells. Interestingly, different cats have different versions of the gene (alleles) that vary in the nucleotide sequence of the binding sites. If the binding site is normal (wild type) it is indicated by a (+) in the table, and this will mean protein 1 or 2 can bind to its site. If the binding site is mutated it will be indicated with a (), and this will mean protein 1 or 2 canngt bind to the DNA. Here are your results: LEVEL OF DOG mRNA 200 50 50 0 50 SAMPLE SITE 1 SITE 2 CAT 1 CAT 2 CAT 3 CAT 4 CAT 5 Things to consider before you begin the online quiz: What is the function of proteins 1 and 2 in the regulation of the DOG gene? (Are they activators, repressors, or some combination of the two?) How do Proteins 1 and 2 affect each other's function? .What might happen to the regulation How many base-pairs of DNA would be present in the binding sites? In the regions marked A and B What is the expected status of Site 1 and Site 2 n Cat #5? (+or-) . Would you predict that Cats #2-5 have less fear of dogs? why or why not?) CULLEGE RULEDExplanation / Answer
1. Proteis 1 and 2 are activators that bind to the regulatory portion of the gene an enhance its transcription
2. Protein 1 and 2 both positively regulate the gene expression. As when both are present and bind to sites 1 and 2, the levels of mRNA are highest, they act cooperatively and positively regulate each other's function
3. It depends on whether this additional protein is an activator or repressor. If its another activator or enhancer it will further increase the gene expression
5. In cat 5, either of the site is mutated.
6. higher the expression higher is the fear of dogs. Cat 4 has no fear of dogs. and cats 2,3,5 have less fear of dogs. this is because of low level of DOG mRNA which would result in low levels of DOG protein giving cats the observed phenotype.
