Approximately one in every 3200 US Caucasian newborns have a disease called cystic fibrosis in which the lung tissue is clogged with thick mucus. Afflicted people rarely live to reproductive age. A mutation on a single gene causes cystic fibrosis and it is a recessive allele (c). The normal condition is dominant (C). a) Calculate the frequency of the c allele in the population (i.e. b) Now calculate the frequency of the C allele (i.e., p). c Now that you know the frequency of p and q, calculate the expected frequencies of all three genotypes in the population (CC, Cc, & cc). 1 out of every 3200 US Caucasian have CF disease. That is 100/3200 % i.e 0.03125 % have CF.

The frequency of C allele be p and that of c allele is q. Remember 2 basic formulas, p + q = 1 and p2 + 2pq + q2 = 1.

a. As q2 = 0.03125% = 0.0003125, q = 0.01767.

b. p = 1 - q = 1 - 0.01767 = 0.98233

Therefore frequency of C is 98.233 % and c is 1.767%.

c. Frequency of Cc is 2pq = 2 X 0.98233 X 0.01767 = 0.03472 i.e 3.472%.

Frequency of CC is p2 = 0.96497 i.e 96.496%.

Frequency of cc is q2 = 0.000312 i.e 0.0312%.

d. Heterozygous for recessive is Cc. The frequency of Cc is 3.472%. Therefore in 10000 people 347 people will be Heterozygous recessive.

e. The frequency of c allele will increase as the C allele wont be able to survive diarrhea and hence C allele frequency will decrease. As frequency is relative term drecrease in frequency of C allele means increase in frequency of c allele.
