Question
5. You are comparing the levels of protein Z from two liver cell cultures. Culture 1 came from a healthy individual, while culture 2 came from a patient with liver cancer. You found that culture 2 contains high levels of protein Z as compared to culture 1. Further analysis showed that the cancer patient contained less miRNA, called miRNA442, compared to the healthy patient. The sequence of miRNA442 is 5'CCUGACGU3" (A) Circle the type of treatment you would recommend to the patient with liver cancer: Antagomir for miRNA442 Artificial miRNA442 (B) Write out the sequence of the mRNA for protein Z that will bind to your treatment (5 to 3')Explanation / Answer
Answer A:- miRNA are the small non coding RNA molecules that are rrsprespons for RNA silencing and post transcriptional regulation of gene expression.
Now if miRNA442 is less in the patient, then Artificial miRNA442 can be introduced into the patient so that it binds to its target protein Z and silences it. So the correct approach for treatment would be to introduce artificial miRNA442 into patient.
Answer B:- The sequence for mRNA of Protein Z will be as follows:
5'UCGTCUGG3'.
As you can see it is a reverse complement of the miRNA sequence. This is important so that miRNA can bind to it.
