Question

4. You are a new researcher in your lab and want to make your mark by determining the function of unknown human genes. You are looking through unannotated parts of the human genome and find two intriguing genes: A and B. With only the DNA sequences, you are unable to determine the function of the genes. However, what can you conclude about the cellular location of the gene products from the following in (A) The final four amino acids of gene A are (in order) lysine, aspartic acid, glutamic acid and leucine (followed by the C-terminus). (B) After the starting methionine, the next five amino acids of gene B are either lysine or arginine.

Explanation / Answer

Answer A - Given sequence is called KDEL sequence and is used for retention of protein in the lumen of endoplasmic reticulum. Therefore gene product will be observed in lumen of ER.

Answer B - Such a signal is used for protein import to the mitochondria. Therefore such gene product will be found in mitochondria.
