1b) Transconjugants are selected by plating on tetracycline antibiotic. The transconjugants containing tetracycline resstance genes will be able to grow in the antibiotic but the E. coli lacking tetr genes will be unable to grow in the antibiotic.
2b) Transconjugants are selected by plating them on spectinomycin and kanamycin antibiotic. Transconjugants whose pigment genes have been knocked out can be selected by plating on kanamycin antibiotic.
