Question
7) (5 points) ?nRH is a hypothalamic neuropeptide that controls mammalian reproduction. Total RNA was extracted from the rat hypothalamus tissue and Northern blot analysis was performed with oligo probes complementary to the 5'end of ?nR mRNA to detect this mRNA. Lane 1 shows blot of control in vitro synthesized GnRH mRNA (560nt). Lane 2 shows blot of the total RNA extract. 4300 nt Explain why lane 2 displays a large smear with several major bands showing above the mRNA band. What is the mechanism that generates these additional bands? D RNA ?Explanation / Answer
This difference is appeard because lane one only have control sample in which only GnRNA is present. But lane 2 shows the sample which is isolated from cells. In cells where is different kind of RNA are present which are involved in the formation of various different proteins.
Due to the multiple kind of RNA the banding pattern is increased because every RNA have different size and Molecular weight.
In cells various different genes are expressed and produce different different RNA which may or may not be involved in the formation of protein.
Means in simple word we can say that this banding pattern is due to multiple kind of RNA which are present in the sample.
