Doctor Octopus has identified the human homolog of the gene of Four-leaf clovers(whose protein is similar to Felix Felicus), For homologous recombination of the gene in the human(children and teenagers), Dr. Octopus must incorporate the original DNA of 4-LC which transcribed primary transcript, not the modified cDNA which is generated from the mRNA of 4-LC by Reverse Transcription.

cDNA is the complementary DNA which is generated from mRNA. mRNA of a species is processed from primary transcript by Post-Transcriptional Modification process. Primary transcript is the product of transcription from DNA. Primary transcript contains both Introns and Exons whereas mRNA contains only Exons. That means if cDNA is being generated from mRNA, that will only contain the DNA parts of Exons, the parts of Introns won't be present on the cDNA but for homologous recombination process, the DNA sequence must same as the homologs. The Introns and exons part both should be present on the DNA, that means there is no possibility of recombination occurrence for the cDNA. Cause of that PCR on cDNA from plant tissue have no need for this experiment.
