Since the stock has the concentration of 100,000 and required is 100 pfu/mL we need 1000 times dilution

We can start as follow

Tube 1= 1mL of stock + 9mL of diluent = 1/10 dilution

Tube 2= 1mL from tube 1 + 9mL of diluent = 1/100 dilution

Take 1mL from the tube 2 and spread it on the sterile broth 9 mL = 1/1000 dilution

So, we expect 100pfu/mL on the broth
